Evaluation Of Essential Oils For Their Activities Against B Burgdorferi Stationary Phase Cultures

To evaluate the possible activity of the essential oils against stationary phase B. burgdorferi, 10% DMSO-soluble essential oils or aqueous suspension of DMSO-insoluble essential oils were added to 100 µL of the seven-day old stationary phase B. burgdorferi culture in 96-well plate to obtain the desired concentrations. In the primary screen, each essential oil was assayed with final concentrations of 0.2% and 0.1% in 96-well plates. Drugs of daptomycin, doxycycline, and cefuroxime were used as control with final concentration of 40 μM. The active hits were checked further with lower concentrations of 0.1% and 0.05% all of the tests mentioned above were run in triplicate. All of the plates were sealed and incubated at 33 °C without shaking for seven days, and 5% CO2 were maintained in the incubator.

In an article published in Antibiotics, a peer-reviewed journal, scientists led by Dr. Ying Zhang identified 10 EOs that have strong activity against stationary phase B. burgdorferi at a low concentration of 0.1%. The study, underwritten by GLA, found that of the 10 EOs, those of garlic, allspice, and Palmarosa were active at even at 0.05% concentration. In addition, cinnamaldehyde, a major ingredient isolated from cinnamon bark, was active against both stationary phase bacteria as well as replicating B. burgdorferi at a 0.02% concentration.

Oils from garlic and several other common herbs and medicinal plants show strong activity against the bacterium that causes Lyme disease, according to a study by researchers at Johns Hopkins Bloomberg School of Public Health. These oils may be especially useful in alleviating Lyme symptoms that persist despite standard antibiotic treatment, the study also suggests.

The study, published October 16 in the journal Antibiotics, included lab-dish tests of 35 essential oils — oils that are pressed from plants or their fruits and contain the plant’s main fragrance, or “essence.” The Bloomberg School researchers found that 10 of these, including oils from garlic cloves, myrrh trees, thyme leaves, cinnamon bark, allspice berries and cumin seeds, showed strong killing activity against dormant and slow-growing “persister” forms of the Lyme disease bacterium.

“We found that these essential oils were even better at killing the ‘persister’ forms of Lyme bacteria than standard Lyme antibiotics,” says study senior author Ying Zhang, MD, PhD, professor in the Department of Molecular Microbiology and Immunology at the Bloomberg School.

Previous studies have shown that fibroblasts provided protection against ceftriaxone . Here we evaluated whether a powerful persister drug combination CefU+ Dox + Dap, which is known to completely eradicate B. burgdorferi aggregated microcolony persisters in vitro could eradicate B. burgdorferi from fibroblast cells. Surprisingly, as shown in Figure 1 and Table 1, the presence of 3T3 fibroblast cells protected the B. burgdorferi from being completely killed not only by single antibiotics doxycycline, cefuroxime, or daptomycin but also by the triple drug combination Cefuroxime+ Doxycycline+ Daptomycin as shown by residual green GFP labeled B. burgdorferi remaining after drug treatment for 7 days. However, we did find that Cefuroxime+ Doxycycline+ Daptomycin combination was more effective in killing B. burgdorferi than any other single antibiotics . Nevertheless, none of them including the triple drug combination Cefuroxime+ Doxycycline+ Daptomycin could completely kill B. burgdorferi as shown by visible spirochetal growth after 21-day subculture .
