Doxycycline , cefuroxime and daptomycin were dissolved in suitable solvents to form 5 mg/ml stock solutions. The antibiotic stocks were filter-sterilized by 0.2 Î¼m filter and stored at â20Â°C. Essential oils were also dissolved in organic solvent dimethyl sulfoxide at 20%, followed by dilution to achieve desired dilutions as previously described .
Evaluation Of Essential Oils For Their Activities Against B Burgdorferi Stationary Phase Cultures
To evaluate the possible activity of the essential oils against stationary phase B. burgdorferi, 10% DMSO-soluble essential oils or aqueous suspension of DMSO-insoluble essential oils were added to 100 ÂµL of the seven-day old stationary phase B. burgdorferi culture in 96-well plate to obtain the desired concentrations. In the primary screen, each essential oil was assayed with final concentrations of 0.2% and 0.1% in 96-well plates. Drugs of daptomycin, doxycycline, and cefuroxime were used as control with final concentration of 40 Î¼M. The active hits were checked further with lower concentrations of 0.1% and 0.05% all of the tests mentioned above were run in triplicate. All of the plates were sealed and incubated at 33 Â°C without shaking for seven days, and 5% CO2 were maintained in the incubator.

All culture medium was filter-sterilized by 0.2 µm filter. Cultures were incubated in sterile 50 ml conical tubes in microaerophilic incubator without antibiotics. After incubation for 7 days, 1 ml stationary-phase B. burgdorferi culture was transferred into a 96-well plate for evaluation of potential anti-persister activity of essential oils .

Persister forms of B. burgdorferi, the bacterium that causes Lyme disease, are dormant or slow-growing, and tolerant of antibiotic treatment. Its not clear yet whether persister bacteria, immune dysfunction, or some combination of the two is responsible for post-treatment Lyme disease syndrome , in which patients treated with antibiotics continue to suffer symptoms.
The search for novel compounds to kill persister bacteria has led to the discovery that essential oils , aromatic compounds produced by plants, may be promising. In an article published in Antibiotics, a peer-reviewed journal, scientists led by Dr. Ying Zhang identified 10 EOs that have strong activity against stationary phase B. burgdorferi at a low concentration of 0.1%. The study, underwritten by GLA, found that of the 10 EOs, those of garlic, allspice, and Palmarosa were active at even at 0.05% concentration. In addition, cinnamaldehyde, a major ingredient isolated from cinnamon bark, was active against both stationary phase bacteria as well as replicating B. burgdorferi at a 0.02% concentration.

Oils from garlic and several other common herbs and medicinal plants show strong activity against the bacterium that causes Lyme disease, according to a study by researchers at Johns Hopkins Bloomberg School of Public Health. These oils may be especially useful in alleviating Lyme symptoms that persist despite standard antibiotic treatment, the study also suggests.
The study, published October 16 in the journal Antibiotics, included lab-dish tests of 35 essential oils — oils that are pressed from plants or their fruits and contain the plant’s main fragrance, or “essence.” The Bloomberg School researchers found that 10 of these, including oils from garlic cloves, myrrh trees, thyme leaves, cinnamon bark, allspice berries and cumin seeds, showed strong killing activity against dormant and slow-growing “persister” forms of the Lyme disease bacterium.
“We found that these essential oils were even better at killing the ‘persister’ forms of Lyme bacteria than standard Lyme antibiotics,” says study senior author Ying Zhang, MD, PhD, professor in the Department of Molecular Microbiology and Immunology at the Bloomberg School.
Chelation Therapy For Lyme Disease
Its done using a type of medication called a chelator or chelating agent. This medication binds to metals in the bloodstream, collecting them into a compound that can be processed by your kidneys and released in urine.
Chelation therapy is an effective treatment for a buildup of heavy metals. But theres no evidence that heavy metals contribute to Lyme disease, and chelation therapy wont treat the underlying infection.

Fibroblast Cells Protect B Burgdorferi From Being Killed By Different Antibiotics Including Drug Combination Cefuroxime+ Doxycycline+ Daptomycin
Previous studies have shown that fibroblasts provided protection against ceftriaxone . Here we evaluated whether a powerful persister drug combination CefU+ Dox + Dap, which is known to completely eradicate B. burgdorferi aggregated microcolony persisters in vitro could eradicate B. burgdorferi from fibroblast cells. Surprisingly, as shown in Figure 1 and Table 1, the presence of 3T3 fibroblast cells protected the B. burgdorferi from being completely killed not only by single antibiotics doxycycline, cefuroxime, or daptomycin but also by the triple drug combination Cefuroxime+ Doxycycline+ Daptomycin as shown by residual green GFP labeled B. burgdorferi remaining after drug treatment for 7 days. However, we did find that Cefuroxime+ Doxycycline+ Daptomycin combination was more effective in killing B. burgdorferi than any other single antibiotics . Nevertheless, none of them including the triple drug combination Cefuroxime+ Doxycycline+ Daptomycin could completely kill B. burgdorferi as shown by visible spirochetal growth after 21-day subculture .
