Subculture Studies To Evaluate The Activity Of Natural Product Extracts Against Stationary Phase B Burgdorferi

To confirm the activity of the natural product extracts in eradicating the stationary phase B. burgdorferi cells, we performed subculture studies as previously described . We further tested the top active natural product extracts to ascertain if they could eradicate stationary phase B. burgdorferi cells at 1 or 0.5% by subculture after the treatment . Treatment with 1% Cryptolepis sanguinolenta extract caused no regrowth in the subculture study . However, the other natural product extracts including Polygonum cuspidatum, Artemisia annua, Juglans nigra, and Uncaria tomentosa could not eradicate B. burgdorferi stationary phase cells as many spirochetes were still visible after 21-day subculture . At 0.5%, all the natural product extracts treated samples grew back after 21-day subculture , however, only one of the three Cryptolepis sanguinolenta extract treated samples grew back. This indicates that 0.5% Cryptolepis sanguinolenta extract still has strong activity and could almost eradicate the stationary phase B. burgdorferi cells. By contrast, the clinically used antibiotics doxycycline and cefuroxime at clinically relevant concentration could not sterilize the B. burgdorferi stationary phase culture, since spirochetes were visible after 21-day subculture .

When Inflammation Becomes Chronic

Imagine if the acute inflammatory process was going on in tissues throughout your body and it never resolved. Thats exactly what happens with the chronic inflammation associated with chronic Lyme disease.

The driving force is bacteria invading cells. Its not an all-out invasion like you would find with an acute infection, such as pneumonia, however. Its an insidious migration of bacteria into tissues throughout the body in the brain, heart, joints, muscles, kidneys, intestines, everywhere.

When Lyme bacteria enter the bloodstream from a tick bite, WBCs gobble up the bacteria in an attempt to get rid of them. The bacteria, however, have sophisticated ways of staying alive inside the WBCs. Instead of eradicating the bacteria, the WBCs unintentionally become transport vehicles to distribute bacteria to tissues throughout the body.

On arrival, the bacteria surface and infect other cells. Its not as much a deluge of invasion as it is a random sprinkling. Tissues throughout the body become peppered with cells that have been infected with bacteria.

The Lyme bacteria, Borrelia burgdorferi, are intracellular, which means they infect and live inside cells. The cells provide nutrients to make new bacteria as well as protection from the immune system. Once the bacteria have used up the resources a cell has to offer, they surface and infect other cells.

    • Wear closed shoes or boots, long-sleeved shirts, and long pants. Tuck your pant legs into your shoes or boots to prevent ticks from crawling up your legs.
    • Use an insect repellent containing 10% to 30% DEET .

    • Don’t sit on the ground outside.

    If you use an insect repellent containing DEET, follow the directions on the product’s label and don’t overapply it. Place DEET on shirt collars and sleeves and pant cuffs, and only use it directly on exposed areas of skin. Be sure to wash it off when you go back indoors.

    Because the activity of antibiotics against non-growing B. burgdorferi is not always correlated with their activity against growing bacteria , we therefore determined the MICs of these natural product extracts against the replicating B. burgdorferi as described previously . The MIC values of some natural product extracts such as Artemisia annua, Juglans nigra, Uncaria tomentosa were quite high for growing B. burgdorferi, despite their strong activity against the non-growing stationary phase B. burgdorferi cells .     Proponents of banderol and samento recommend them as alternatives to antibiotics, which are the standard treatments for Lyme disease. It’s said that these herbal remedies may help patients steer clear of the side effects sometimes associated with antibiotic use, such as gastrointestinal distress.

    Supporters also say that banderol and samento aid in the treatment of Lyme disease by reducing chronic inflammation.

    In addition, banderol and samento are sometimes used to control post-treatment Lyme disease syndrome . In people with PTLDS, symptoms linger long after they’ve completed a course of antibiotics. These symptoms include fatigue, muscle and joint pain, sleep disruption, and mood changes.

    At this point, however, theres very little evidence to back up claims that banderol or samento can help treat Lyme disease. There is some research out there showing that a combination of samento and banderol may help knock out Borrelia burgdorferi, the bacterium responsible for Lyme disease.     Like many in the Hudson Valley, Lyme disease has turned the Callahans’ lives upside down. Both of their children are afflicted with the illness, which has proven to be a powerful foe for their youngsters to fight.

    “Lyme disease has impacted our lives on and off for five years, and daily for over a year,” Laura Callahan said, who lives in Milton, Ulster County. “My son Ryan was unable to attend school for most of last year.     Dr. Richard I. Horowitz, author of the New York Times best-seller, “Why Can’t I Get Better? Solving the Mystery of Lyme and Chronic Disease,” and medical director of the Hudson Valley Healing Arts Center in Hyde Park contends that the method used to calculate the 300,000 figure is flawed. It doesn’t take into account the fact that Lyme is a multi-systemic malady.

    Under-reporting and misdiagnosis in children is due in part to the fact they often aren’t able to describe their symptoms accurately.

    How all this plays out is a function of the health of cells as much as the aggressiveness of the bacteria: Cells that are weak from being chronically stressed are more vulnerable to invasion by bacteria.

    A healthy person with healthy cells who is bitten by a tick may not have much in the way of acute symptoms and may never develop chronic symptoms. It doesnt mean, however, that the bacteria have been eradicated from the body. They can stay dormant in tissues for a lifetime.

    All it takes is a shift in the health of cells for dormant bacteria to surface and invade more cells. The immune system scrambles to keep a lid on things as bacteria erupt and infect other cells. Dead cells break apart and create debris. Macrophages secrete acid and free radicals to break down debris. Reinforcements are called in. More acid. More free radicals. More collateral damage. More debris. Flow of nutrients and oxygen is obstructed and cells are weakened even further.

    The invasion intensifies as more cells are infected. Along with inflicting injury to cells in tissues, bacteria infect and kill WBCs. In addition, bacteria throw the immune system off balance by disrupting the cytokines the immune system uses to coordinate the resistance. As the process spirals out of control, the immune system loses the capacity to keep microbes in check.

    Before long, tissues throughout the body start to look like a war zone. Cells suffer. Symptoms result. It becomes a never-ending cycle of misery.
