Detecting Lyme In Hiding

Since borrelia flourishes in ticks, why not see if ticks can be used to find live spirochetes hiding in people? A bizarre new way to look for infection, xenodiagnosis, seems like it might be a possibility. This technique consists of placing uninfected ticks on peoples arms, letting them feed for a few days, removing them, and seeing if they now contain live borrelia. In a small pilot study of sixteen people with PTLDS, one tick acquired B. burgdorferi DNA from a persons blood. That doesnt unequivocally prove that the person was still infected because the ticks could be taking up bits of DNA from leftover dead bacteria. A confirmation would require taking that tick, putting it on a mouse, and that mouse becoming infected. This was not the case for the one positive tickit was not infectious .

Azlocillin Comes Out On Top

The drug, which is not on the market, was tested in mouse models of Lyme disease at seven-day, 14-day and 21-day intervals and found to eliminate the infection. For the first time, azlocillin was also shown to be effective in killing drug-tolerant forms of B. burgdorferi in lab dishes, indicating that it may work as a therapy for lingering symptoms of Lyme disease.

Pothineni and Rajadas have patented the compound for the treatment of Lyme disease and are working with a company to develop an oral form of the drug. Researchers plan to conduct a clinical trial.

Rajadas is also a professor of bioengineering and therapeutic sciences at the University of California-San Francisco.

Other Stanford co-authors are Hari-Hara S. K. Potula, PhD, senior research scientist postdoctoral scholars Aditya Ambati, PhD, and Venkata Mallajosyula, PhD senior research scientist Mohammed Inayathullah, PhD and intern Mohamed Sohail Ahmed.

Screening thousands of drugs, Stanford scientists determined that in mice, azlocillin, an antibiotic approved by the Food and Drug Administration, eliminated the bacteria that causes Lyme disease.

For decades, the routine treatment for Lyme disease has been standard antibiotics, which usually kill off the infection. But for up to 20% of people with the tick-borne illness, the antibiotics dont work, and lingering symptoms of muscle pain, fatigue and cognitive impairment can continue for years sometimes indefinitely.

A new Stanford Medicine study in lab dishes and mice provides evidence that the drug azlocillin completely kills off the disease-causing bacteria Borrelia burgdorferi at the onset of the illness. The study suggests it could also be effective for treating patients infected with drug-tolerant bacteria that may cause lingering symptoms.

This compound is just amazing, said Jayakumar Rajadas, PhD, assistant professor of medicine and director of the Biomaterials and Advanced Drug Delivery Laboratory at the Stanford School of Medicine. It clears the infection without a lot of side effects. We are hoping to repurpose it as an oral treatment for Lyme disease. Rajadas is the senior author of the study, which was published online March 2 in Scientific Reports. The lead author is research associate Venkata Raveendra Pothineni, PhD.

Hunting For Alternative Drug

Frustrated by the lack of treatment options for Lyme disease patients with lingering symptoms, Rajadas and his team began hunting for a better alternative in 2011. In 2016, they published a study in Drug Design, Development and Therapy that listed 20 chemical compounds, from about 4,000, that were most effective at killing the infection in mice. All 20 had been approved by the Food and Drug Administration for various uses. One, for instance, is used to treat alcohol abuse disorder.

In this most recent study, azlocillin, one of the top-20 contenders, was shown to eclipse a total of 7,450 compounds because it is more effective in killing B. burgdorferi and causes fewer side effects. Lyme disease affects more than 300,000 people annually, according to the Centers for Disease Control and Prevention. It can affect various organs, including the brain, skin, heart, joints and nervous system, and cause heart problems and arthritis if untreated. Symptoms include fever, headaches, chills, and muscle and joint pain.

Traditional antibiotics, such as doxycycline, are effective as an early course of treatment for the infection in the majority of patients, but it remains unclear why these drugs fail to treat 10% to 20% of patients, Rajadas said.

Tryptophan For Mood Support

Tryptophan is an amino acid, and like most amino acids, its a building block for proteins. But tryptophan is also used to make the mood-regulating neurotransmitter serotonin. Tryptophan metabolism appears to be altered in Lyme disease, as in other infections , and it has been suggested that tryptophan supplements could be helpful in supporting serotonin production and mood in people with Lyme disease. Taking tryptophan supplements and supplements of other building blocks for neurotransmitters, such as tyrosine and choline, has been proposed for many conditions where the brain is not working optimally. It isnt easy to eat enough supplementary amino acids to affect brain neurotransmitters significantly because neurons tightly regulate the amount of neurotransmitters they make. But there is some evidence that the activated form of tryptophan, 5-HTP, may help support mood regulation . There is not yet evidence of efficacy in treating Lyme disease.

If this were true it would be one of the greatest discoveries in the history of medicine. It also it would be horrific pulsed radio transmission is kind of a fancy way to talk about a radio tower the one that sends sounds to your car radio, for example.

Imagine the power needed to send a radio signal over the air and what would happen if doing so produced mechanical resonance within the cells of the body.

If the Rife machines mechanism of action actually was effective, wed cure cancer when we tune to the classic rock station. More likely, if it did work, wed have LOTS of cancer!

Of course the rife doesnt work. The companies do not offer any evidence. If you click on their positive clinical results it is not clinical research, but a story about the man who invented the machine.

If you have the Lyme rash but havent had it for more than seven days, you may wish to see if you qualify for this trial. The standard test looks for antibodies to the Lyme bacterium in blood. It takes a while for antibodies to show up, so if you dont have antibodies, you dont know whether its just too early to see them. The MENSA approach is to look at the antibodies that white blood cells are beginning to makethis should be a much better indicator of an active infection.

Antibody And Western Blot Tests

Diagnosis typically includes testing for antibodies your body has made in response to Lyme bacteria. If antibiotic treatment is started early, the body may never develop antibodies. Somewhere between a few days and six weeks after a tick bite, blood tests called ELISA, EIA, and IFA can pick up antibodies: The IgM type of antibody is made first and then the IgG type. Antibodies can stick around for months or years, though, so they dont tell you much about when you were infected or whether you are still infected. And unfortunately, the antibodies wont protect you if you get another bite from a borrelia-carrying tick in the future .

Antibody tests are not infallible, and they may come up with both false positive and false negative results. If an antibody test result is positive, standard practice is to do a Western blot or immunoblot test to rule out a false positive result and be more certain that borrelia bacteria are present. The antibody test followed by the Western blot is called a standard two-tiered test .

There appear to be two ways to get better results by stage 2 that are not yet standard practice. One is to do two EIA tests, and the other is to use a test called VlsE or C6. Based on research in children with clinical symptoms of Lyme disease, researchers have suggested that when the STTT is negative but children have Lyme symptoms, additional tests should be carried out .
